Reconstitution as a laboratory procedure
Dissolution arithmetic and nothing more. This page contains no dose, no route and no protocol, and we cannot advise on use of any kind.
The procedure
- Let the vial reach room temperature before opening. Introducing solvent to cold powder drives condensation into it, and moisture is what degrades a lyophilised peptide.
- Add the solvent down the inside wall of the vial, not directly onto the powder. A jet into the cake drives peptide to the air-liquid interface, where it denatures.
- Do not shake. Swirl gently, or set it down and let it dissolve. Some sequences take several minutes; a long or hydrophobic peptide can take considerably longer.
- Look at it. A fully dissolved peptide solution is clear. Persistent cloudiness or visible particulate means it has not gone into solution, and more agitation is not the answer.
- Record the volume you used. Everything below depends on it.
The arithmetic
Concentration is the amount in the vial divided by the volume of solvent you added. It is not affected by anything else.
- 5mg into 1ml5mg/ml
- 5mg into 2ml2.5mg/ml
- 10mg into 1ml10mg/ml
- 10mg into 5ml2mg/ml
For material sold in international units rather than by mass — HCG is the one in this catalogue — the same division applies but the result is units per millilitre. An international unit measures biological activity, not mass, so the two are not interchangeable and cannot be converted without a potency figure specific to the material.
Which solvent
Bacteriostatic water is water for injection with 0.9%% benzyl alcohol as a preservative, which is what permits a vial to be entered more than once. It is the usual choice.
Dilute acetic acid is used for peptides that dissolve poorly in water alone, typically the longer and more hydrophobic sequences.
